MINI PARASEP®
STEP 1:
Sample Preparation
A) Unscrew lid.
B) Add 2.4ml of fixative.
C) Add one drop of surfactant (eg: Apacor Triton X solution)
to the chamber.
Alternatively, use reagent ready Mini Parasep®.
D) Introduce a level scoop of formed faecal sample(equivalent 0.5g) to the fixative using the spoon at the end of the Mini Parasep® filter. For liquid samples, add 2 level scoops (equivalent 1g) to the fixative. Add 1.0ml of Ethyl Acetate to the mixing chamber. Mix in thoroughly with the Mini Parasep® spoon. If the sample is hard, break it up with the end of the spoon.
STEP 2:
Emulsification
A) Seal Mini Parasep® by screwing in the filter/sedimentation cone unit. Vortex or shake to emulsify with the sedimentation cone pointing upwards.
STEP 3:
Centrifugation
A) Invert Mini Parasep® and centrifuge at 1200g for 3 minutes. Mini Parasep® fits all 15ml centrifuge buckets.
STEP 4:
Examination
Open very slowly to avoid aerosol release.
A) Unscrew and discard the filter and mixing tube.
B) Pour off all the liquid above the sediment.
C) Pipette one drop of sediment onto a slide and cover with coverslip.
Alternatively, follow laboratory SOP for slide preparation.
