PARABEADS

To validate both Midi and Mini filters, for both Parasep® and Parasep® SF

CONTENTS PROVIDED IN PACK

1 vial containing 1ml red ParaBeads® solution should be sufficient for testing:
– 10 x Mini Parasep® / Mini Parasep® SF
– 5 x Midi Parasep® / Midi Parasep® SF

The ParaBeads® will sink to the bottom of the vial. In order to ensure that they are fully dispersed, the vial containing ParaBeads® should be agitated as mentioned below:
– either by vortexing for 10 seconds;
– or by shaking vigorously 10 times before use.

N.B. ParaBeads® must not be used with AlcorFix or Ethyl Acetate as it could cause the beads to dissolve. For prefilled AlcorFix tubes, decant fixative into liquid waste, rinse the tube, and replace with water (Mini 3ml, Midi 6ml).

 

 

For both Mini Parasep® and Mini Parasep® SF

1. Once shaken, add 100μl of the ParaBeads® solution to the mixing tube (containing any fixative other than AlcorFix as described above)
NB. Do not add ethyl acetate as this will dissolve the beads.

2. Screw the cap on the mixing tube and shake the fixative and ParaBeads® solution to mix.

3. Unscrew the cap and pipette a 20μl sample of the solution onto a microscope slide for observation at 10x magnification. Count the number of beads in one Field of View (FOV).

4. Then take the Parasep Mixing tube containing the ParaBeads® and screw in the filter assembly to run the Parasep®procedure as per the instructions for use.

5. The ParaBeads® will then be concentrated at the bottom of the sedimentation cone. Pipette a 20μl sample from the pellet for mounting on a slide. Count and compare with previous FOV at 10x magnification. A minimum concentration coefficient of 5x should be observed.

For both Midi Parasep® and Midi Parasep® SF

1. Once shaken, add 200μl of the ParaBeads® solution to the mixing tube (containing any fixative other than AlcorFix as described above)
NB. Do not add ethyl acetate as this will dissolve the beads.

2. Screw the cap on the mixing tube and shake the fixative and ParaBeads® solution to mix.

3. Unscrew the cap and pipette a 20μl sample of the solution onto a microscope slide for observation at 10x magnification. Count the number of beads in one Field of View (FOV).

4. Then take the Parasep Mixing tube containing the ParaBeads® and screw in the filter assembly to run the Parasep®procedure as per the instructions for use.
